Solid surface vitrification of human spermatozoa: Comparision of three cryoprotectants

Abstract

The present study aims to compare effect of cryopreservation medium has components cryoprotectant to different whether the protective effects of semen cryopreservation and decrease sperm death.

          Semen samples collected from 47 normospermic male volunteers. Semen were sperm prepared by Discontinuous density gradient technique and divided into 4 groups. Group 1: sperm preparation (Pre-freeze). Group 2: Sperm freezing with glycerol egg yolk citrate (GEYC). Group 3: Sperm freezing with glycerol citrate (GC). Group 4: Sperm freezing with Human sperm preservation medium (HSPM). The semen quality of these four groups were compere i.e. semen analysis for motility, progressive motility, normal sperm morphology and vitality.

          Significantly decreased in motility, progressive motility, normal sperm morphology and vitality were found in experiment groups compared Pre-freeze group. On the other hand, GEYC and HSPM were motility, progressive motility and vitality increased significantly compared to GC. Furthermore, GEYC was normal sperm morphology increased significantly compared GC and HSPM.

          Cryopreservation with GEYC medium and HSPM medium lead to increased motility, progressive motility and vitality compared GC medium. In GEYC medium is egg yolk component more than normal sperm morphology. Similarly, GEYC medium and HSPM medium appropriate for Solid surface vitrification.
