Abstract

To facilitate genetic diversity preservation and conservation initiatives, poultry semen cryopreservation remains as a critical technology in avian reproduction. Despite notable achievements in assisted reproductive technologies in the species in the past decades, the biochemical and physiological traits of chicken semen pose unique challenges for cryopreservation. This review examined the factors influencing the quality, fertility, and freezability of cryopreserved chicken semen while emphasizing on the role of proteins, antioxidants, and cryoprotectants like ethylene glycol and dimethylacetamide in maintaining sperm integrity. The fundamentals of cryopreservation, including important procedures like collection, dilution, cooling, and thawing were also discussed alongside advances in current techniques such as slow freezing and vitrification, and novel approaches like microfluidics and nanoparticle-based cryoprotection. The effects of lipid peroxidation and oxidative stress on sperm structure and DNA integrity during freezing was also elaborated, suggesting new cryoprotectants and antioxidants as mitigating measures. Breed-specific variations and environmental factors are also explored, underscoring the need for customized strategies to maximize cryopreservation results. The analysis also looked at promising technologies for improving cryopreservation outcomes such as artificial sperm reservoirs. This article also highlights the significance of ongoing research to improve the dependability and accessibility of semen cryopreservation for both commercial and conservation purposes, while addressing contemporary issues like standardization and ethical considerations. In conclusion, this comprehensive review of existing knowledge and emerging trends provides a roadmap for transformative research and development breakthroughs in poultry semen reproductive technologies.
