Abstract
Objective: To study the three method of preparation of artificial cerebrospinal fluid and observe the bacterial contamination
after performing each technique. Materials and methods: Preparation lymphocyte cell from EDTA blood
with high lymphocyte high lymphocyte discarded after HLA typing and apply lymphoproliferation assay ,three
methods wererinsed bysolution and centrifuged with any rpm. To get lymphocyte cell.The lymphocytes cell from
three methods were counted by three technicians and mixed to neutrophils cell according to case study.The lymphocytes
and neutrophils were smeared for differentiation. Result: The comparison of three methods to preparation
lymphocytes cell showed that three techniques yielded are no significant difference for the lymphocyte cell count (pvalue
≤ 0.05) but the lymphoproliferation assay provided no bacterial contamination and easy to preparation in one
day. Conclusion: Artificial cerebrospinal fluid prepared by provided higher lymphocyte count with no bacterial contamination
better than the two methods and distinct morphology in Wright stain. So this technique could be used as learning and teaching material.
