The study of the effect of soap on reducing the hardness of formalin-fixed paraffin embedded (FFPE) tissue

Objective : Application of soapwater to soften the hard paraffin-embedded tissue block
Method : Samples from myoma uteri and thyroid (n=240) are processed through histopathological preparation and then soaking in soap water at pH 7 and pH 10 for 1, 2, 3, and 4 minutes per each block and preparing slides. Assess the softness and quality of H&E-stained slides.
Results : Soaking paraffin-embedded tissue in soapy water for 3 minutes results in best tissue softness (10 out of 21 blocks (47.6%) in pH 7 and 9 out of 14 blocks (64.3%) in pH 10). Poor quality of H&E-stained slides (10 out of 10 blocks (100%)) is related to soaking in pH 10 for 3 or 4 minutes.
Conclusion : Soaking paraffin-embedded tissue in soapy water at pH 7 for 3 minutes gives the best tissue softness and quality of H&E-stained slides.
