NAT2-diplotyping by Real-time PCR
Keywords:
NAT2-diplotyping, Isoniazid, Real-time PCR, PCR, AcetylatorAbstract
N-acetyltransferase 2 enzyme (NAT2) plays an important role in the metabolizing of Isoniazid, an antituberculosis drug. Individuals having a slow acetylator phenotype were found 8.8 times more at risk to be TB drug-induced liver injury compared to intermediate and fast acetylator ones. The acetylator phenotype can be interpreted from NAT2-diplotyping and used for Isoniazid-dose adjustment. This study aimed to develop a Real-time PCR for NAT2-diplotyping to replace the previous PCR for routine service. Optimization of primer/probe concentration and temperature/time condition was done with known phase-analysis NAT2-diplotyping DNA samples. The DNA detection range of this new method was 5–150 ng/μL. Method verification of the Real-time PCR for NAT2-diplotyping by comparing it with the PCR method in 138 samples. The result has shown that NAT2-diplotype in all samples from both methods was 100% consistent. In conclusion, the novel real-time NAT2-diplotyping which provides faster results and is more convenient, can replace the previous PCR for clinical routine service.
