บทคัดย่อ

       การศึกษาครั้งนี้ได้คัดเลือกเปปไทด์จากฐานข้อมูลเปปไทด์ต้านจุลินทรีย์จำนวน 9 เส้น ที่มีขนาด 6-25 กรดอะมิโนมีประจุสุทธิเป็นบวก และ 40-83% hydrophobicity นำเปปไทด์ทั้งหมดไปสังเคราะห์และทดสอบฤทธิ์ยับยั้งการเจริญของเชื้อ Staphylococcus aureus ATCC 25923, methicillin-resistant Staphylococcus aureus (MRSA) DMST 20635, MRSA DMST 20637 และ MRSA DMST 20654 พบว่าเปปไทด์ KLKLLLLLKLK สามารถยับยั้งเชื้อ S. aureus ทั้งหมด จากนั้นทำการดัดแปลงเปปไทด์เส้นนี้ ทำให้ได้เปปไทด์จำนวน 12 เส้น ที่มีค่าของ %hydrophobicity และประจุสุทธิแตกต่างจากเดิม ในการทดสอบฤทธิ์ยับยั้งแบคทีเรียของเปปไทด์สังเคราะห์ 12 เส้น พบว่าเปปไทด์ KLKLKLKLKLK, LLLLLLKLK และ LLLLLLK มีประสิทธิภาพยับยั้งเชื้อ S. aureus ATCC 25923, MRSA DMST 20635 และ MRSA DMST 20654 ได้สูงสุด โดยมีค่า IC50 27.5-62.5 ไมโครกรัมต่อมิลลิลิตร นอกจากนี้ผลการวิเคราะห์ matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF MS) พบว่าทุกสายพันธุ์ของเชื้อ S. aureus มี Peptide barcode แตกต่างกัน จากผลการทดลองครั้งนี้แสดงให้เห็นว่า S. aureus ที่ดื้อยาเมทิซิลลินต่างกันมีผลต่อ Peptide barcode ต่างกัน และการตอบสนองต่อเปปไทด์สังเคราะห์ก็แตกต่างกันด้วย อย่างไรก็ตามกลไกการทำงานของเปปไทด์ทั้ง 3 เส้น ที่มีฤทธิ์ยับยั้งเชื้อ MRSA ควรจะนำไปศึกษาเพื่อพัฒนาเป็นยารักษาโรค
